Cold Shock Susceptibility of Boar
نویسندگان
چکیده
F OUR trials were conducted with boar spermatozoa to determine the effects of dilution rate, seminal plasma content and extender composition on susceptibility to cold shock and the development of resistance to cold during 30 C incubation. Acrosome morphology and sperm motility were used to assess damage by cold shock. The susceptibility of boar spermatozoa to cold shock increased as the dilution rate increased from 1:2 to 1:6 or 1:10. In addition, spermatozoa diluted to 1:6 or 1:10 before incubation were more susceptible to cold shock than spermatozoa diluted 1:2 before incubation and then rediluted to 1:6 or 1:10 after incubation and immediately before cold shock. Spermatozoa that were washed before incubation were more resistant to cold shock after 5-hr. incubation than after i-hr. incubation, indicating that the acquisition of resistance to cold shock during incubation was an inherent property of the spermatozoa. However, seminal plasma provided boar spermatozoa with additional protection against cold shock. The replacement of seminal plasma with Tris-lactose extender containing 2 to 4% casein protected sperm acrosomes against cold shock but did not reduce the adverse effect of cold shock on sperm motility.

Comparison of Motility, Acrosome, Viability and ATP of Boar Sperm with or without Cold Shock Resistance in Liquid Semen at 17°C and 4°C, and Frozen-thawed Semen
This study was designed to analyze boar sperm to compare motility, acrosome morphology, viability and ATP by various preservation methods between Duroc boar A with cold shock resistance sperm and Duroc boar B with cold shock sensitivity sperm. Semen volume, sperm concentration, motility and normal acrosome between Duroc boar A and B did not show any differences within 2 h after collection. Ther...
متن کاملImmunocytochemical detection of acrosomal damage following cold shock: loss of acrosin from the acrosomal region of ram, bull and boar spermatozoa.
Samples of ram, bull and boar semen were subjected to cold shock and then stained using an indirect immunocytochemical method for detecting acrosin. It was found that the shock treatment abolished staining of the acrosomal region in all but a very few cells. This finding was interpreted as a great loss of proacrosin or acrosin from damaged acrosomes. Using a fluorescent label in the system, spe...
متن کاملInteractions of egg yolk lipoprotein fraction with boar spermatozoa assessed with a fluorescent membrane probe.
The interactions of a fluorescent membrane probe, 1-anilinonaphthalene-8-sulfonic acid (1,8-ANS), with boar spermatozoa were followed through the use of lipoprotein fraction of ostrich egg yolk (LPFo). Semen samples, extended in Kortowo 3 (K3) extender, were supplemented with 2% or 5% LPFo and stored for 3h at 16 degrees C. Additionally, cold shock-treated spermatozoa (1h at 4 degrees C) were s...
متن کاملExogenous Individual Lecithin-Phospholipids (Phosphatidylcholine and Phosphatidylglycerol) Cannot Prevent the Oxidative Stress Imposed by Cryopreservation of Boar Sperm
For artificial insemination (AI), boar semen is prepared in rather simple extenders, kept at 16-20°C and usually used in liquid form for up to 3 days from extension. Routine freezing of boar semen is occasionally used for gene banking, replacement of lost genetics or long-distance transport [1]. The reason behind this meagre use of freezing is the low tolerance of the ejaculated boar sperm to c...
متن کاملZinc-binding proteins from boar seminal plasma -- isolation, biochemical characteristics and influence on spermatozoa stored at 4°C.
Affinity chromatography on Chelating Sepharose Fast Flow Gel-Zn(2+) was used for fractionation of boar seminal plasma proteins. Approximately 30% of total boar seminal plasma proteins showed affinity for zinc ions (ZnBP fraction). Native electrophoresis (PAGE) of ZnBP revealed six protein fractions which separated into 27 bands under denaturing conditions (SDS/PAGE). Two-dimensional electrophor...
